Indian researchers find rare gene mutation linked to recurrent neurological decline in kids

Indian researchers find rare gene mutation linked to recurrent neurological decline in kids

New Delhi, Nov 27: A team of Indian researchers has identified a rare USP18 gene mutation linked to recurrent neurological decline in children.

The rare USP18 gene mutation offers crucial insights into a neurological disorder previously documented in only 11 cases worldwide and now reported for the first time in India, according to the team from Indira Gandhi Institute of Child Health, Bangalore, who conducted the research in collaboration with Ramjas College, University of Delhi, and Redcliffe Labs.

Pseudo-TORCH syndrome type 2 is a very rare inherited condition that affects how a child’s brain grows and functions. Children with this disorder often show severe neurological symptoms that resemble congenital infections, but without any actual infection.

The USP18 gene usually helps regulate the body’s immune response, preventing excessive inflammation. When this gene does not work correctly, the body’s defence system becomes overactive and begins to harm the brain.

The research, published in the journal Clinical Dysmorphology, documented a previously unreported variant, c.358C>T (p.Pro120Ser), expanding clinical understanding of Pseudo-TORCH syndrome type 2.

“The discovery reinforces the power of clinical intuition backed by advanced genetic testing. For years, we treated symptoms without a definitive answer, but identifying this novel USP18 mutation has transformed not just the diagnosis, but the child’s future,” Dr. Vykuntaraju K. Gowda, Department of Pediatric Neurology, Indira Gandhi Institute of Child Health (IGICH, told IANS.

“The finding will help us avoid unnecessary treatments, provide precise therapy, and most importantly, guide families through informed genetic counselling. Our research shows how timely genetic insights can change the course of rare neurological disorders, offering hope where answers were once unknown,” Gowda added.

The research was initiated with an 11-year-old girl who began experiencing symptoms from infancy, including repeated episodes of febrile encephalopathy, that is, fever-linked unconsciousness, seizures, delayed development, and a small head size.

Over the years, her scans also showed increasing calcium deposits in different parts of the brain.

To understand the underlying cause of these recurring neurological problems, advanced genetic testing was recommended. Through a focused DNA test called exome sequencing with mitochondrial genome sequencing, the team identified a previously unknown change in the USP18 gene, offering clarity after years of uncertainty.

The newly identified mutation alters the structure of the USP18 protein, weakening its ability to control inflammation. This overactive immune response can explain the child’s repeated episodes of fever-triggered neurological decline.

Understanding this connection is essential because it helps doctors recognise early warning signs, avoid unnecessary treatments for infections, and focus instead on monitoring and managing conditions linked to immune overactivation.

“The case is also the first documented case of USP18-related disease presenting with recurrent febrile encephalopathy,” Dr Himani Pandey, one of the researchers.

The research highlights the importance of early genetic testing in children with unexplained neurological symptoms and opens the door for more targeted approaches to care in the future.

Aga Ruhullah’s counter to Deputy CM’s “limelight” remark

Srinagar, Nov 27: Srinagar MP Aga Ruhullah Mehdi on Thursday said that his political stance is driven by principles and not positions of power, a day after Deputy Chief Minister, Surinder Choudhary advised him to raise his voice in Parliament and stop seeking the limelight.

Speaking with reporters, Aga Ruhullah said that people who lack discipline, principle, honesty, and loyalty in their politics are not even worthy of a response.

“The Deputy CM left the PDP, and yet he advises me about principles and party. My fight is for principles, not for the chair. I have not changed the party and principles for the chair,” he said.

He said that two years ago, the Deputy CM was not in the National Conference, and sometimes aligned with the BJP, sometimes with the PDP. “Wherever he gets the chair and opportunity, he goes there,” Ruhullah said.

He also said that such people have no principles, no values, and no direction in their lives. “My politics and my efforts are based on principles and values. That is why, if I have expressed disagreement with the party, it is based on principles, and not for the chair,” he said—(KNO)

J&K Govt blacklists LMES IT LLP, bars firm from all future recruitment contracts

J&K Govt blacklists LMES IT LLP, bars firm from all future recruitment contracts

Srinagar, Nov 27: The Jammu and Kashmir government has permanently blacklisted M/s LMES IT LLP and its director, Maharaj Krishan Wali, in connection with the 2020 Firemen and Firemen Drivers recruitment scam, barring them from participating in any future recruitment-related tenders in the Union Territory.

The decision, issued through Government Order No. 1540-JK(GAD) of 2025 by the General Administration Department (Vigilance), follows a multi-layered inquiry into alleged irregularities during the recruitment conducted by the Fire and Emergency Services Department.

According to the order, the government had appointed a committee in December 2022 to probe the recruitment process. The committee submitted its findings in February 2024, after which the matter was forwarded to the Anti-Corruption Bureau (ACB). The ACB, upon verification, registered FIR No. 01/2025 at its Central Jammu Police Station earlier this year.

Investigations revealed that LMES IT LLP failed to meet mandatory technical and eligibility criteria laid out in the tender. The firm had no prior experience in large-scale recruitment for police, paramilitary or state forces—an essential requirement. It was also discovered that the company was incorporated only in November 2017, after splitting from Energy and Software Pvt Ltd, and did not qualify as a reputed recruitment vendor.

Authorities noted that LMES relied heavily on financial documents of another firm to meet the tender criteria and submitted only one balance sheet of its own. Despite being ranked L-2 in the bidding process, the firm was awarded the contract while the L-1 bidder’s case was left unresolved—pointing to procedural violations.

The government order also highlights that LMES director Maharaj Krishan Wali had previously worked with Timing Technologies Pvt Ltd, a firm whose recruitment contract had been cancelled in 2018. Despite this conflict of interest, LMES was allowed to participate and subsequently secure the Firemen recruitment contract, indicating that the vendor was extended undue favour.

Acting on the Home Department’s recommendations under Rule 151 of the General Financial Rules (GFR) 2017, the UT administration has now sanctioned the firm’s permanent blacklisting from all recruitment-related bidding processes.
